Overview

PRDM16 divergent transcript (PRDM16-DT) is a long non-coding RNA located on chromosome 1p36.32[4] that encodes a novel small protein. This protein regulates alternative splicing of the CHEK2 gene, competitively interacts with HNRNPA2B1, and suppresses cancer metastasis and chemoresistance by promoting E-cadherin secretion and inhibiting fibroblast activation[1][2]. PRDM16-DT is highly expressed in astrocytes in the human brain and is downregulated in neurodegenerative conditions such as Alzheimer’s disease[6]. Low expression of PRDM16-DT has been identified as a biomarker for poor prognosis in several cancers, and its transcript, LINC00982, is transcribed divergently from PRDM16[3][6]. Cimicifugoside H-1 has been identified as a small molecule inducer of PRDM16-DT via FOXP3, offering a potential therapeutic approach for colorectal cancer[2].
