Pre–T-cell antigen receptor alpha (PTCRA) is a single-pass type I transmembrane protein and a member of the immunoglobulin superfamily expressed in immature thymocytes. It forms a key part of the pre-T-cell receptor (pre-TCR) complex by covalently associating with the T-cell receptor beta (TCRβ) chain and the CD3 signaling complex; this complex is essential for early T-cell development in the thymus. PTCRA’s primary function is to permit survival, proliferation, and differentiation of thymocytes with successful TCRβ rearrangement, regulate TCRβ allelic exclusion, and allow αβ T cell lineage commitment. The receptor operates in a ligand-independent, self-oligomerizing fashion, with signaling dependent on the associated CD3 complex. Deficiency or mutation of PTCRA leads to developmental arrest of αβ T cells and immunodeficiency, while its dysregulation can contribute to certain immune or lymphoproliferative disorders. The gene encoding PTCRA is also known as pT-alpha and is conserved in amniotes. There are no drugs targeting PTCRA, but its function is vital in basic T-cell biology and immunodeficiency research.
