Pre-B-cell leukemia transcription factor 2 (PBX2) is a ubiquitously expressed transcription factor belonging to the TALE (Three Amino acid Loop Extension) homeobox gene family. PBX2 functions as a transcriptional activator by binding to specific DNA sequences (notably 5'-ATCAATCAA-3'), activating gene expression such as PF4 when dimerized with cofactors like MEIS1. PBX2 is implicated in cell fate specification, proliferation, and developmental regulation, and has links to several diseases, most prominently leukemia and congenital cardiac defects. PBX2 can serve as a putative biomarker for cancer, but no direct therapeutic agents nor established mechanisms of drug action are currently described for targeting this protein directly in clinical practice. PBX2 acts in cellular pathways through complex formation with other transcription factors, influencing the gene regulatory networks essential for normal development and disease progression.
