Pre-mRNA-splicing factor CWC22 homolog (CWC22) is a highly conserved protein-coding gene required for pre-mRNA splicing as a component of the spliceosome. It enables RNA binding and participates in processing capped intron-containing pre-mRNA, functioning mainly in the cytosol and nuclear specks. CWC22 is essential for exon junction complex (EJC) assembly and thereby for nonsense-mediated mRNA decay, acting by recruiting and escorting proteins such as EIF4A3 to the splicing machinery. CWC22 forms part of both the major (U2-type) and minor spliceosome complexes and interacts with multiple nuclear proteins. Disease associations are limited, with reports relating to hard palate cancer and root caries, but evidence for it as a therapeutic target is absent in current literature.
