Pre-mRNA-splicing factor ISY1 homolog (ISY1) is a spliceosome-associated protein involved in pre-mRNA splicing within the nucleus, functioning as part of the U2-type catalytic spliceosome complex. ISY1 has RNA binding activity and is necessary for the proper processing and splicing of nuclear RNAs. It also participates in selective microRNA biogenesis during embryonic stem cell differentiation and the transition between pluripotency states. Notably, ISY1 directly interacts with and enhances the endonuclease activity of APE1, a key enzyme in base excision DNA repair (BER), and its expression is upregulated under oxidative stress, supporting cell survival in the presence of DNA damaging agents. ISY1’s spliceosomal function is evolutionarily conserved from yeast to human, acting as a non-snRNA protein component involved in nuclear mRNA splicing. There are no known drugs, direct biomarkers, or significant safety concerns associated with ISY1; its relevance is primarily basic biological and not as a therapeutic target.
