Preferentially Expressed Antigen in Melanoma (PRAME) is a cancer/testis antigen highly expressed in various cancers, including melanoma and leukemia, but minimally expressed in normal adult tissues (except testis). It functions as a corepressor of retinoic acid receptor signaling, inhibiting differentiation and apoptosis. PRAME is an attractive target for cancer immunotherapy due to its restricted expression pattern and immunogenicity, and it also has diagnostic utility in distinguishing malignant melanoma from benign nevi.
PRAME is targeted by immunotherapies (peptide vaccines, adoptive T-cell therapies) that stimulate cytotoxic T lymphocytes to recognize and kill PRAME-expressing cells.
