Overview

The PRAME ALY peptide–HLA-A*02 complex is a specific peptide-major histocompatibility complex (pMHC) presented on the surface of cancer cells. PRAME (Preferentially expressed Antigen in Melanoma) is a cancer-testis antigen that is highly expressed in various malignancies, including melanoma, sarcoma, and ovarian cancer, while remaining largely absent in healthy adult tissues except for immune-privileged sites like the testes (UniProt P78395; PubMed 32814731). The ALYVDSLFFL peptide is a specific 10-amino acid sequence derived from the PRAME protein that is processed and loaded onto HLA-A*02:01 molecules for presentation to T cells (Immatics, 2023). This complex serves as a highly specific target for T-cell receptor (TCR)-based therapies, such as TCR-engineered T cells (TCR-T) and TCR-bispecific molecules (PubMed 35413154). By targeting this pMHC, these therapies can selectively direct the immune system to recognize and eliminate PRAME-positive tumor cells (Medigene, 2022). Clinical development of agents targeting this complex, such as IMA203, is ongoing, with a focus on maximizing anti-tumor efficacy while minimizing off-target reactivity (ClinicalTrials.gov NCT03686124).
