Overview

Prefoldin subunit 2 (PFDN2) is a beta subunit of the prefoldin complex, a heterohexameric molecular chaperone that binds and stabilizes newly synthesized polypeptides—primarily actin and tubulin—to ensure proper folding and prevent aggregation. The prefoldin complex facilitates the transfer of these polypeptides to the cytosolic chaperonin TCP-1/TRiC/CCT for final folding steps, crucial for cytoskeletal maintenance and protein homeostasis. PFDN2, along with other subunits, is evolutionarily conserved and important for cellular processes such as mitosis, neuronal development, and cellular stress responses. Alterations in PFDN2 expression or function have been associated with disease states including various cancers and neurodegenerative conditions due to its role in protein folding and prevention of aggregation. There is currently no evidence that PFDN2 is a direct pharmacological target, nor are there established biomarkers or safety concerns specific to its modulation in therapy.
