Pregnancy-specific beta-1-glycoprotein 3 (PSG3) is a member of the pregnancy-specific glycoprotein family, which are major secreted proteins produced by placental syncytiotrophoblasts and released into the maternal circulation during pregnancy. PSG3 is one of several closely related glycoproteins (PSG1-PSG11 in humans) within the immunoglobulin superfamily. They are primarily thought to play a role in promoting immune tolerance of the mother toward the fetus, supporting placental development, and modulating inflammation. PSG3 is not classically regarded as a druggable receptor, enzyme, transporter, or canonical immune checkpoint target, and no approved drugs are known to selectively target this molecule. Its major relevance clinically is as a biomarker for pregnancy status and potentially for abnormal pregnancy conditions when measured in maternal blood. PSG3 should not be confused with the scaffolding protein supramodule "PSG" (PDZ3-SH3-GK) found in postsynaptic proteins, as this is unrelated at the molecular, functional, and biological level.
