Overview

PRELI domain-containing 1 pseudogene 6 (PRELID1P6) is a long noncoding RNA pseudogene closely related to the PRELID1 gene. Although classified as a pseudogene, PRELID1P6 is upregulated in glioma and correlates with poor patient survival. Functional studies demonstrate that PRELID1P6 promotes glioma cell proliferation and inhibits apoptosis by stabilizing the protein hnRNPH1, leading to increased TRF2 expression, altered splicing, and activation of the oncogenic Akt/mTOR signaling pathway. Knockdown of PRELID1P6 blocks cell cycle progression and enhances apoptosis in glioma cells. Its role appears to be regulatory, functioning at the RNA level by interacting with RNA-binding proteins (notably hnRNPH1) and potentially competing with microRNAs (such as miR-1825)[1]. PRELID1P6 is not a receptor, enzyme, or druggable target in the classic sense, but it may serve as a novel prognostic biomarker and a candidate for research in the context of glioma biology and therapeutic resistance[1].

Mechanism of action

Drugs: Inhibition of the Akt/mTOR signaling pathway (MK2206). Endogenous: Stabilization of hnRNPH1, activation of Akt/mTOR pathway, promotion of alternative splicing, and suppression of apoptosis.

03

Biological functions

Regulation of cell proliferationRegulation of apoptosisModulation of protein stability (through inhibition of ubiquitin-mediated degradation of hnRNPH1)Potential regulation of gene expression via interaction with RNA-binding proteins and possibly as a miRNA sponge
04

Disease associations

Cancer (specifically glioma)
05

Safety considerations

Not applicable; as a pseudogene, PRELID1P6 itself is not a drug target. However, its upregulation is associated with adverse prognosis in glioma

Interacting drugs

MK2206
07

Biomarkers

PRELID1P6 expression is a proposed biomarker for glioma prognosis and may predict response to therapies targeting the Akt/mTOR pathway
