PRELI domain containing protein 3B (PRELID3B) is a member of the PRELI/MSF1 (Slowmo/Ups) domain-containing lipid transfer protein family, localized to the mitochondrial intermembrane space[1][6]. PRELID3B forms a complex with TRIAP1, acting as a specific mitochondrial phosphatidylserine (PS) transporter that shuttles PS across the mitochondrial intermembrane space[1][6]. Structurally, it has an internal lipid-binding pocket with a flexible entrance, which is important for its lipid specificity and transfer function[1]. PRELID3B and its associated proteins are evolutionarily conserved and critical for maintaining proper mitochondrial lipid homeostasis[1][6]. While alterations in PRELI-like proteins are associated with disruptions in lipid metabolism and have been correlated with diseases such as cancer, PRELID3B itself is not characterized as a direct therapeutic target nor is it currently the target of known drugs[6].
