PreQ1 synthase is an enzyme that catalyzes the four-electron reduction of the cyano group of 7-cyano-7-deazaguanine (preQ0) to an amine group, producing preQ1, an intermediate in the biosynthesis of queuosine[7]. Queuosine is a hypermodified nucleoside incorporated into the wobble position of certain tRNAs, enhancing translational fidelity and potentially influencing bacterial phenotype and virulence[3][5]. PreQ1 synthase is classified as an oxidoreductase due to its role in reductive chemical transformations. Although the queuosine biosynthetic pathway is essential in bacteria, and thus enzymes like preQ1 synthase are candidates for antibacterial target validation, no clinically used drugs target this enzyme at present.
