Presynaptic cholinergic receptors refer to a heterogeneous group of receptors located on the distal ends of cholinergic neurons that modulate the synthesis and release of acetylcholine (ACh) (Wonnacott, 1997). The primary components include muscarinic autoreceptors, predominantly M2 and M4 subtypes, and nicotinic receptors, such as alpha-7 and alpha-4-beta-2 subtypes (Zhang et al., 2002; UniProt). Muscarinic autoreceptors typically provide negative feedback, inhibiting further ACh release when synaptic concentrations are high, whereas presynaptic nicotinic receptors often facilitate ACh release (StatPearls, 2023). These receptors are critical for maintaining cholinergic homeostasis and are significant in the pathology of neurodegenerative conditions like Alzheimer's disease, where the loss of presynaptic terminals leads to a profound cholinergic deficit (Francis et al., 1999). Pharmacological targeting of these receptors aims to enhance cholinergic transmission, either by blocking inhibitory autoreceptors or by stimulating facilitatory nicotinic receptors (PubMed, 2021).
