Overview

The presynaptic neuromuscular junction is the terminal end of a motor neuron axon that forms a synapse with a skeletal muscle fiber. Its primary function is to release acetylcholine (ACh) into the synaptic cleft upon arrival of an action potential. This release is dependent on calcium influx, which triggers the fusion of ACh-containing vesicles with the presynaptic membrane. The released ACh then diffuses across the synaptic cleft and binds to nicotinic acetylcholine receptors on the muscle fiber, initiating muscle contraction. Dysregulation of presynaptic function can lead to neuromuscular disorders and is a target for various drugs and toxins.
