Prickle planar cell polarity protein 2 (PRICKLE2) is a cytoplasmic signaling and scaffold protein critical for neuronal development, where it regulates planar cell polarity and microtubule bundling, particularly at the axon initial segment (AIS) in the brain[1][2]. PRICKLE2 is essential for correct synaptic development, proper post-synaptic density organization, and the maturation of axonal structures[1][2][3]. Mutations or loss of function in PRICKLE2 disrupts neuronal morphology, reduces dendritic branching and synaptic density, and is causally linked to progressive myoclonic epilepsy type 5 as well as autism spectrum disorder–like behaviors and neurodevelopmental deficits[3]. PRICKLE2 does not act as a classical drug target such as a receptor or enzyme; there are no known pharmacological modulators or interacting drugs[3]. Its main significance is as a regulatory node in neuronal polarity maintenance and central nervous system cytoarchitecture[1][2][3].
