Overview

PRKAG2 antisense RNA 1 (PRKAG2-AS1) is a long non-coding RNA transcribed antisense to the PRKAG2 gene, implicated in the regulation of gene expression at the transcriptional or post-transcriptional level. It is overexpressed in multiple cancer types, including hepatocellular carcinoma, colon adenocarcinoma, advanced prostate cancer, and esophageal squamous cell carcinoma. High PRKAG2-AS1 expression correlates with poor prognosis and increased malignancy in cancer and is a promising prognostic biomarker as well as a potential therapeutic target due to its regulation of oncogenic pathways, particularly via miR-502-3p/BICD2 signaling

Mechanism of action

Silencing or inhibiting PRKAG2-AS1 leads to suppression of oncogenic traits (proliferation, migration, invasion) in cancer cells, likely via regulation of the miR-502-3p/BICD2 axis and modulation of downstream genes involved in EMT

03

Biological functions

Regulation of gene expression (specifically, PRKAG2)Promotes cancer cell proliferation, migration, and invasionFunctions as a competing endogenous RNA, binding microRNAs such as miR-502-3p and regulating the BICD2 axis in hepatocellular carcinomaModulation of epithelial-to-mesenchymal transition (EMT) markers
04

Disease associations

Cancer (notably hepatocellular carcinoma, colon adenocarcinoma, prostate cancer, esophageal squamous cell carcinoma)
05

Safety considerations

No known safety concerns or therapeutic challenges specifically documented for directly targeting PRKAG2-AS1; clinical experience is limited as this is an emerging target
06

Interacting drugs

None currently known or reported
07

Biomarkers

PRKAG2-AS1 expression in tissue may serve as a prognostic biomarker for disease-free and overall survival in hepatocellular carcinoma and possibly other cancers
