Overview

PRKAR1B antisense RNA 2 (PRKAR1B-AS2) is a long noncoding RNA transcribed antisense to the protein-coding gene PRKAR1B on chromosome 7. It is overexpressed in ovarian cancer, where it promotes tumor cell proliferation, migration, invasion, and chemoresistance (notably to cisplatin and alpelisib) by modulating the PI3K/AKT/mTOR pathway, primarily through direct interaction with PI3K-110α. Knockdown of PRKAR1B-AS2 inhibits these processes and enhances chemosensitivity in both in vitro and animal xenograft models, highlighting its potential as both a therapeutic target and a biomarker for cancer progression and drug resistance

Mechanism of action

Targeting PRKAR1B-AS2 (e.g., by small interfering RNA) leads to downregulation of PI3K-110α and inhibition of PI3K/AKT/mTOR pathway signaling, enhancing sensitivity to cisplatin and alpelisib in ovarian cancer models
