PRKG1 antisense RNA 1 (PRKG1-AS1) is a long non-coding RNA predominantly localized in the cell nucleus, which regulates the adjacent DKK1 gene through epigenetic processes including methylation and modulation of EZH2 protein. PRKG1-AS1 is implicated in cancer biology: its overexpression promotes cell proliferation, migration, and poor prognosis in lung adenocarcinoma, while in gastric cancer, it interacts with another lncRNA, UC.145, and helps determine cancer cell signaling output via DKK1 and Wnt signaling pathways. Knockdown of PRKG1-AS1 reduces cell migration, proliferation, and apoptosis rates in vitro, revealing its function in controlling cell fate decisions relevant to oncogenesis. PRKG1-AS1 is currently studied as a prognostic biomarker in cancers but is not directly targeted by approved medicines or experimental drugs as of date.
