Overview

The expression of Interleukin-6 (IL-6), Interleukin-8 (IL-8), and Interleukin-1 beta (IL-1β) represents a critical readout of the pro-inflammatory state in the human body. These cytokines are primarily produced by macrophages, monocytes, and endothelial cells in response to various stimuli, including pathogens and tissue damage (NCBI PMC7291910). IL-6 is a pleiotropic cytokine that drives the acute phase response and T-cell differentiation (UniProt P05231), while IL-8 (CXCL8) acts as a major chemoattractant for neutrophils to sites of inflammation (UniProt P10145). IL-1β is a potent pyrogen and inflammatory mediator that requires processing by the NLRP3 inflammasome for its activation and secretion (UniProt P01584). Elevated levels of these cytokines are hallmark features of "cytokine storms" seen in conditions like sepsis, CAR-T cell therapy complications, and severe viral infections (PubMed PMID: 32433836). Therapeutic strategies often focus on reducing the expression or activity of these molecules to mitigate tissue damage in autoimmune and autoinflammatory diseases using agents like tocilizumab or canakinumab (FDA/PubChem).

Mechanism of action

Inhibition of cytokine signaling through direct neutralization of the cytokine, blockade of its cognate receptor, or suppression of upstream transcriptional pathways such as NF-κB and JAK/STAT (PubMed PMID: 25135167).
