Overview

Pro-inflammatory cytokine receptors are a broad class of transmembrane proteins that serve as critical mediators of the inflammatory response by binding to signaling molecules such as interleukins and tumor necrosis factors [StatPearls: Cytokines]. These receptors, which include the IL-1, IL-6, and TNF receptor families, initiate intracellular signaling cascades—most notably the NF-κB and JAK/STAT pathways—that regulate immune cell activation, proliferation, and survival [Nature Reviews Immunology: Cytokine receptors]. Dysregulation or chronic overactivation of these receptor systems is a hallmark of numerous pathological states, including rheumatoid arthritis, psoriasis, and systemic inflammatory response syndrome [PubMed: PMC4124030]. Therapeutic intervention often involves the use of monoclonal antibodies or decoy receptors designed to block ligand-receptor interactions, thereby dampening the inflammatory cascade [PubMed: PMC7147065].
