Pro-inflammatory factor expression refers to the upregulation and secretion of molecules—primarily cytokines—that promote inflammation. These factors are crucial mediators in the immune response, orchestrating the activation, recruitment, and proliferation of immune cells in response to infection, injury, or other inflammatory stimuli. The term encompasses a broad set of gene products whose expression is tightly regulated at both transcriptional and post-transcriptional levels. Main pro-inflammatory factors are cytokines such as: Interleukins: IL-1 (α/β), IL-2, IL-6, IL-12, IL-17, IL-18, Tumor Necrosis Factor alpha (TNF-α), Interferon gamma (IFN-γ). Other contributors include chemokines (e.g., IL-8), granulocyte-macrophage colony-stimulating factor (GM-CSF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), transforming growth factor beta (TGF-beta) under certain contexts. Pro-inflammatory factors are produced predominantly by: Activated macrophages, Dendritic cells, T helper 1 cells (Th1). Expression is controlled by several signaling pathways: NF-kB pathway, MAPK pathway, JAK/STAT pathway. Dysregulated pro-inflammatory factor expression can lead to pathological conditions such as autoimmune diseases or cytokine storm syndromes.
