Overview

Pro-melanin-concentrating hormone (PMCH) is a precursor polypeptide primarily expressed in the hypothalamus, which is proteolytically processed to produce several neuropeptides, including melanin-concentrating hormone (MCH), neuropeptide-glutamic acid-isoleucine (NEI), and neuropeptide-glycine-glutamic acid (NGE). MCH, the best-studied product, functions as an orexigenic (hunger-promoting) neuropeptide and plays critical roles in the regulation of feeding behavior, energy balance, sleep-wake cycles, and potentially reproductive or maternal behaviors. The PMCH gene is primarily expressed in the lateral hypothalamic area, with processed peptides acting via specific G protein-coupled receptors (MCHR1 and MCHR2 in humans and some species). Dysfunction or abnormal signaling in this pathway has been implicated in metabolic disorders such as obesity, as well as in the regulation of mood and sleep. PMCH itself is not considered a direct therapeutic target; pharmacological interventions to date focus on the peptide products (primarily MCH) or their receptors[1][2][3].
