Proapoptotic nucleolar protein 1 (PANO1) is a nucleolar protein that induces apoptosis and participates in tumor suppression pathways. It achieves this mainly by modulating and stabilizing the tumor suppressor protein CDKN2A/p14ARF, thus protecting it from degradation. By doing so, PANO1 may contribute to enhanced cell cycle arrest and apoptosis in response to cellular stress, and it has been postulated as a potential tumor suppressor[5]. PANO1 localizes primarily to the nucleolus and is involved in several regulatory processes, notably the negative regulation of proteasomal ubiquitin-dependent protein catabolic process and positive regulation of apoptotic pathways[5]. It is also known by the alias "pre-mRNA-splicing factor cwc22-like" in some databases, likely based on gene orthology, but its primary established function is apoptosis induction and tumor suppression rather than splicing[4][5][6]. There are no current drugs targeting PANO1 directly, and it is not yet a clinically established biomarker. The major research focus is its role in cell death and cancer biology.
