Overview

Probable ATP-dependent RNA helicase DDX60-like (DDX60L) is a member of the DExD/H-box helicase family and is an interferon-stimulated gene product with demonstrated antiviral activity, particularly against hepatitis C virus[2][3]. DDX60L is structurally and functionally related to DDX60 but is distinct in its ability to restrict hepatitis C virus (HCV) replication in hepatic cell models. DDX60L is upregulated upon viral infection and interferon stimulation, localizes predominantly in the cytoplasm, and acts as an effector of the innate immune response by directly inhibiting viral RNA replication, with limited effect on RNA translation or stability. It has only a moderate impact on RIG-I-dependent innate immune signaling, suggesting a direct antiviral effector role rather than an indirect regulatory function[2][3][5]. Excessive overexpression of DDX60L in vitro may be cytopathic, but under physiological conditions, it serves as part of the interferon-mediated antiviral defense system. There are currently no known drugs officially reported to modulate DDX60L, nor is it established as a biomarker for patient selection or safety monitoring[2][3][5].
