Probable G protein-coupled receptor 156 (GPR156) is an orphan G protein-coupled receptor belonging to the class C, metabotropic glutamate receptor subfamily. It contains seven transmembrane domains and is expressed ubiquitously, with particularly high expression in fetal brain. GPR156 exhibits high constitutive activity and shows strong coupling with Gi/Go family G proteins. Its primary recognized biological role is in the regulation of hair cell orientation in the mechanosensory organs of the inner ear, where it is essential for auditory function and implicated in congenital hearing loss. Despite some sequence homology to GABA_B receptor subunits, GPR156 lacks a functional response to GABA_B ligands and is considered an orphan receptor. Structural studies reveal a unique dimeric organization and suggest that its activity may be regulated by local phospholipid composition. No validated drug interactions, actionable biomarkers, or specific safety concerns are reported in the current literature for GPR156.
