Overview

Probable G protein-coupled receptor 45 (GPR45) is an orphan G protein-coupled receptor predominantly expressed in the central nervous system—including the hypothalamus, cortex, hippocampus, and brain stem—as well as in the testis[2]. GPR45 is a class A, seven-transmembrane domain receptor whose endogenous ligand and precise physiological signaling partners remain unknown[2][3]. Disruption of Gpr45 in mice leads to increased adiposity, hepatic steatosis, and glucose intolerance, largely due to decreased expression of pro-opiomelanocortin (POMC) in the hypothalamus and reduced energy expenditure mediated via the JAK2/STAT3 signaling pathway[2]. GPR45 is of increasing research interest as a potential drug target for disorders of energy balance, including obesity, but is not yet the target of any approved therapies[2][3].

Mechanism of action

Not established for GPR45-specific drugs. Conceptually, potential agonists could increase energy expenditure and POMC expression by engaging GPR45 and modulating the JAK2/STAT3 pathway[2].
