Overview

PRSS50 (Probable threonine protease PRSS50; also known as testis-specific protease 50 or TSP50) is a protease predominantly expressed in the testes, with highest expression during spermatocyte and spermatid development[1][3][5]. It is implicated in the regulation of sperm morphogenesis by contributing to centrosome function and sperm tail formation. PRSS50 acts as a threonine endopeptidase and is required for normal differentiation and structure of sperm, as knockout studies in mice demonstrate male infertility with multiple sperm abnormalities. In addition to its reproductive role, PRSS50 is ectopically upregulated in several malignancies, where it may promote proliferation, migration, and invasion of cancer cells by activating the NFκB pathway through the degradation of IκB[1]. There is no evidence that PRSS50 is currently a therapeutic target, and no known drugs interact with this protein. Its primary clinical relevance is as a gene required for male fertility and, potentially, as a cancer-associated gene due to its expression in several tumors and involvement in cell proliferation and differentiation pathways[1][3][5].
