Overview

Procyanidins are a class of polyphenolic compounds, specifically oligomeric or polymeric flavan-3-ols, that belong to the broader category of proanthocyanidins or condensed tannins. They are prevalent in various plant-based foods and beverages, including cocoa, apples, grapes, and red wine, and are well-regarded for their potent antioxidant and anti-inflammatory properties (PMID: 21950645). In a therapeutic context, procyanidins are considered bioactive compounds or nutraceuticals rather than biological targets like receptors or enzymes; instead, they exert their effects by interacting with and modulating a wide array of proteins and signaling pathways. Research indicates their potential in reducing the risk of cardiovascular disease by improving endothelial function and inhibiting LDL oxidation, as well as providing neuroprotective effects by mitigating oxidative stress (PMID: 24733362). Despite their biological potential, procyanidins face significant pharmacological challenges, primarily due to their large molecular size and complexity, which lead to poor intestinal absorption and rapid microbial metabolism in the gut (PMID: 22617655).

Mechanism of action

Procyanidins act as free radical scavengers and modulate various molecular targets rather than being targets themselves; they inhibit enzymes such as angiotensin-converting enzyme (ACE) and cyclooxygenase (COX), and they modulate signaling pathways including NF-κB and MAPK.
