Profilin-2 (PFN2) is a member of the profilin family of small actin-binding proteins that regulate cell cytoskeleton dynamics by binding actin monomers, affecting their polymerization and reorganization. PFN2 also interacts with polyproline-rich proteins and phosphoinositides, modulating endocytosis, neurotransmitter release, and vesicle trafficking in neurons, where it is predominantly expressed. PFN2 plays a role in iron metabolism by affecting iron pools and homeostasis, as revealed by knockout models. Although PFN2 is essential for several cellular processes, especially within the CNS, it is not currently considered a druggable therapeutic target or receptor, and no selective modulators are available for clinical or research use[1][2][3].
