Overview

Progestagen-associated endometrial protein (PAEP), also known as glycodelin or placental protein 14 (PP14), is a secreted glycoprotein of the lipocalin superfamily principally expressed in the human reproductive tract, particularly the secretory endometrium, decidua, and placenta[1][2][3]. PAEP plays essential roles in reproduction, including modulation of immune responses (creating maternal tolerance to the fetus), regulation of endometrial receptivity, and control of key fertilization steps via several glycosylated isoforms (glycodelin-A, -S, -F, -C), each with unique functions in sperm-egg interaction and immune regulation[2][3]. It has been implicated in the pathology of several reproductive diseases, including infertility, endometriosis, and recurrent implantation failure, as well as in the progression of various cancers, where its overexpression may enhance cell proliferation, migration, and tumor aggressiveness—particularly in melanoma[1][3]. PAEP is regarded as a key biomarker for endometrial receptivity and may be targeted for diagnostic or potential therapeutic purposes in oncology and reproductive medicine[1][2].
