Overview

Progestin and adipoQ receptor family member 5 (PAQR5), also known as membrane progesterone receptor gamma (mPRγ), is a seven-transmembrane receptor belonging to the membrane progestin receptor subfamily within the progestin and adipoQ receptor (PAQR) family[1][3][4]. PAQR5 is expressed primarily in the plasma membrane of various tissues, with particular abundance in the kidney and regulation observed in reproductive and several other tissues. It mediates rapid, non-genomic signaling in response to progesterone, likely via G(i) protein-coupled pathways, and is implicated in oocyte maturation and broader cell signaling cascades. In clear cell renal cell carcinoma, PAQR5 expression is suppressed—often by promoter methylation—and is associated with poor prognosis, suggesting a tumor-suppressive role, while its modulation and downregulation have also been observed in other cancers. Currently, PAQR5 is not a direct drug target, but its potential as a prognostic biomarker is under active investigation[2][4].
