CD274 messenger RNA (mRNA) is the primary transcript of the CD274 gene, which encodes the Programmed cell death 1 ligand 1 (PD-L1) protein. PD-L1 is a critical transmembrane protein that functions as an immune checkpoint by binding to the PD-1 receptor on T cells, thereby inhibiting T-cell activation and promoting self-tolerance (Source: NCBI Gene, ID: 29126). In the context of oncology, many tumors overexpress CD274 mRNA to produce high levels of PD-L1 protein, allowing them to evade the host immune system (Source: PubMed, PMID: 30410170). While traditional immunotherapies utilize monoclonal antibodies to block the PD-L1 protein, targeting the CD274 mRNA itself using RNA interference (RNAi) or antisense oligonucleotides is an emerging therapeutic approach designed to prevent protein production at the source (Source: PubMed, PMID: 33230236). This strategy aims to achieve more complete silencing of the PD-L1 pathway and may be effective in patients who are resistant to protein-targeting agents.
