Overview

Programmed cell death 1 ligand 2 (PD-L2) is a type I transmembrane protein that functions as an immune checkpoint receptor ligand. It plays a critical role in the negative regulation of the adaptive immune response by binding to its receptor, programmed cell death protein 1 (PD-1), on T cells. This interaction inhibits T-cell activation and proliferation, contributing to immune tolerance and modulation of inflammatory responses. Compared to PD-L1, PD-L2 has a higher binding affinity to PD-1. It is expressed on antigen-presenting cells, certain T cells, and B cells. Its expression is regulated by cytokines such as IL-4, GM-CSF, and interferons. PD-L2 plays a role in tumor immunity and is a target for immunotherapies.
