Overview

PD-L1 is a transmembrane protein that plays a crucial role in suppressing the adaptive immune system. By binding to its receptor PD-1 on T cells, PD-L1 transmits an inhibitory signal, reducing T-cell proliferation and cytokine production. This pathway is important for preventing autoimmunity but can be exploited by tumors to evade immune surveillance, making it a major target for cancer immunotherapy.

Mechanism of action

Inhibition of PD-1 signaling by blocking PD-L1 binding, leading to enhanced T-cell activation and anti-tumor immunity. Recruitment of phosphatases such as SHP-2 via the Immunoreceptor Tyrosine-Based Switch Motif (ITSM) in PD-1 after ligand binding.
