Overview

Programmed cell death protein 2 (PDCD2) is a highly conserved nuclear zinc finger protein expressed in diverse tissues and required for the maintenance and proliferation of stem cells and fast-dividing cells. PDCD2 contains a MYND-type zinc finger domain, facilitating protein-protein interactions, notably with co-repressors and transcriptional regulators. It participates in ribosome maturation, mRNP complex formation, and controls RNA processing for select transposable elements. PDCD2 expression is tightly regulated; repressed by BCL6 and upregulated in malignancies, particularly acute leukemia, where its levels predict disease relapse. Functionally, it is indispensable for embryonic and hematopoietic stem cell maintenance, and its knockdown impairs proliferation of cancer cells in vitro, indicating its promise as a therapeutic target. However, due to its fundamental biological roles, strategies to modulate PDCD2 must be carefully considered to avoid disrupting normal cell growth and stem cell populations.
