Overview

Programmed cell death protein 5 (PDCD5) is a highly conserved regulatory protein originally identified by its ability to accelerate apoptosis in various cell types[4][7]. PDCD5 is expressed in both the cytoplasm and nucleus; during apoptosis, its level increases and it translocates to the nucleus[4]. Structurally, it features a compact helical core and flexible termini[1][2]. PDCD5 interacts with cellular pathways central to apoptotic regulation—including acting as a co-activator of p53 in response to DNA damage—and is implicated in stabilizing SERCA2a during cardiac hypertrophy, suggesting protective feedback against excessive cardiac growth[5][6]. It also binds the TRiC/CCT chaperonin complex in its open state, indicating a potential role in proteostasis and possibly neurodegenerative disorders[2]. PDCD5 is considered a promising target in oncology and cardiology, though there are no known direct approved drugs or clinical inhibitors against it as of now.
