Overview

Programmed cell death protein 6 (PDCD6, also known as ALG-2) is a calcium-binding protein in the penta-EF-hand family that acts as an intracellular calcium sensor and adapter. Calcium binding triggers conformational changes allowing PDCD6 to modulate interactions between diverse protein partners involved in apoptosis, ER-Golgi transport, endosomal biogenesis, and membrane repair. PDCD6 participates in T cell receptor-, Fas-, and glucocorticoid-induced programmed cell death, although evidence from knockout mouse models suggests functional redundancy. PDCD6 also interacts with multiple signaling proteins (e.g., ASK1, Fas receptor, ANXA11) and regulates complexes in the ESCRT pathway, vesicular trafficking, and potentially angiogenesis or viral protein distribution. Disease associations include reported roles in cancer (ocular melanoma) and potentially in infection, with pleiotropic functions in various cellular compartments[1][2][3][5][6].
