Overview

A progranulin modulator refers to any compound or agent that alters the levels or activity of progranulin (PGRN), a multifunctional glycoprotein involved in diverse biological processes including inflammation, neuroprotection, cell proliferation, and lysosomal function. Modulators can either increase (agonists/activators) or decrease (antagonists/inhibitors) PGRN expression or activity. PGRN deficiency is associated with neurodegenerative diseases, while overexpression is associated with certain cancers, creating a complex therapeutic landscape.

02

Mechanism of action

Modulation of progranulin expression or activity; can involve increasing or decreasing PGRN levels, influencing proteolytic cleavage, or targeting interacting proteins/receptors such as EphA2, Sortilin, or TNFR1/2.
