Progression-associated lncRNA in breast cancer (lncRNA-PRLB) is a long non-coding RNA upregulated in breast cancer tissues and cell lines. It promotes cancer cell proliferation, metastasis, and resistance to chemotherapy (notably 5-fluorouracil and paclitaxel) by regulating the stability and expression of SIRT1, a NAD+-dependent deacetylase. lncRNA-PRLB acts by sequestering miR-4766-5p, a microRNA that would otherwise inhibit SIRT1 expression. High levels of lncRNA-PRLB are associated with worse clinical outcomes in breast cancer patients and serve as a potential prognostic biomarker. Although it is not currently a direct therapeutic target, its role in modulating SIRT1 signaling and cancer biology may offer future therapeutic opportunities.
Not applicable as there are no direct drugs specifically targeting lncRNA-PRLB described. The lncRNA-PRLB itself inhibits miR-4766-5p, which normally inhibits SIRT1. Thus, lncRNA-PRLB upregulates SIRT1 protein by "sponging" miR-4766-5p and stabilizing SIRT1, leading to increased proliferation, metastasis, and chemoresistance of breast cancer cells.
