Prohibitin (PHB) refers to a highly conserved family of proteins found in eukaryotes. In humans, it is encoded by the PHB gene located on chromosome 17q21. There are two main types: Prohibitin-1 (PHB1) and Prohibitin-2 (PHB2), which form a large heteromeric complex primarily in the inner mitochondrial membrane but are also present in the nucleus and plasma membrane. Prohibitins play multifaceted roles across several cellular processes including energy metabolism, cell proliferation, apoptosis, senescence, signal transduction, and transcriptional control. Mutations or altered expression of prohibitin have been linked with various diseases including cancer and neurodegenerative diseases.
