Prohibitin pseudogene 5 (PHBP5) is a processed pseudogene located on human chromosome 6q25, sharing high nucleotide sequence identity (over 90%) with its parental gene prohibitin (PHB1). While PHBP5 does not encode a protein, its RNA acts as a long noncoding natural antisense transcript, able to bind and form RNA-RNA duplexes with PHB1 mRNA. This interaction can stabilize PHB1 mRNA, increase PHB1 expression, and thereby potentially promote cancer cell proliferation and progression, as shown in esophageal squamous cell carcinoma. PHBP5 is sometimes confused with PHB1, but its distinct role is post-transcriptional regulation of PHB1, and not as a therapeutic target itself.
