The prokaryotic 30S ribosomal subunit is the smaller component of the prokaryotic 70S ribosome, consisting of a single 16S ribosomal RNA molecule (~1542 nucleotides) and 21 ribosomal proteins (S1-S21). It is essential in translation: responsible for recognizing and decoding the mRNA codon through base-pairing with the tRNA anticodon, ensuring translational fidelity, and guiding the initiator tRNA to the correct start site on the mRNA by binding the Shine–Dalgarno sequence. The 30S subunit also plays a crucial role in coordinating the assembly of the complete ribosome and the translocation of tRNA and mRNA during protein synthesis. This subunit is the primary target of many clinically important antibiotics, including aminoglycosides and tetracyclines, which exert their antibacterial effects by interfering with decoding and translocation processes on the ribosome. Structural insights into the 30S subunit have been foundational for understanding mechanisms of antibiotic action, translation accuracy, and evolution of ribosomal function.
