Overview

Prolargin (PRELP) is a small leucine-rich repeat proteoglycan present in the extracellular matrix of connective tissues. It anchors basement membranes to the underlying connective tissue by binding type I collagen (basement membranes) and type II collagen (cartilage), as well as the proteoglycan perlecan. Its N-terminal domain is rich in proline and arginine and binds glycosaminoglycans, mediating cell adhesion and inhibiting osteoclast differentiation. PRELP variants have been implicated in certain disease contexts (e.g., Hutchinson-Gilford progeria, bladder carcinoma in situ), but are not considered direct drug targets.
