Overview

Proliferation-associated 2G4 pseudogene 4 (PA2G4P4), also known as PA2G4L1, is a pseudogene located on chromosome 3, included in the long intergenic non-coding RNA 886 (LINC00886) locus, and shares sequence similarity with the protein-coding gene PA2G4 (encoding ErbB3-binding protein 1, EBP1)[1]. Unlike its parental gene, PA2G4P4 does not code for a protein but is a long non-coding RNA with regulatory functions. Recent studies have shown that PA2G4P4 is significantly overexpressed in bladder cancer tissues and cell lines. Knockdown of PA2G4P4 inhibits proliferation and migration of bladder cancer cells and increases apoptosis, highlighting its potential oncogenic role in bladder cancer. PA2G4P4 transcript distribution correlates closely with that of the parental EBP1 protein in bladder epithelium and urothelial carcinomas. Although it affects cancer cell characteristics, PA2G4P4 is not a protein, receptor, or canonical drug target, but it may play a role in regulating expression and possibly isoform switching of the parental PA2G4 gene[1][2]. Thus, it is not considered a classical therapeutic target (such as a receptor or enzyme), but may represent a novel biomarker or RNA-based therapeutic candidate in oncology, particularly in bladder cancer.
