Overview

PROSER1 is a chromatin-associated protein that regulates DNA demethylation during neurodevelopment by scaffolding and stabilizing complexes containing TET family DNA demethylases and OGT, a glycosyltransferase[1][3]. It promotes O-GlcNAcylation and stability of TET2, modulates recruitment of TET enzymes to chromatin, and affects histone modifications and enhancer activity[1][3]. Loss of PROSER1 leads to altered DNA methylation patterns and gene expression changes, particularly at CpG islands and enhancers, which can result in neurodevelopmental defects[1]. PROSER1 is highly conserved across mammals, with general cytoplasmic expression and enrichment in tissues such as lymph, embryonic tissue, thymus, and uterus[4][5]. While essential for proper epigenetic regulation and development, it is not currently considered a therapeutic target, and no interacting drugs are documented.
