Proline and serine-rich protein 2 (PROSER2) is an uncharacterized protein encoded by the PROSER2 gene, also known as C10orf47, located at chromosome 10p14. Recent research has identified PROSER2 as a membrane- and cytosol-localized protein; its expression is higher in primary tumor tissues than in metastases for pancreatic ductal adenocarcinoma (PDAC). In functional assays, overexpression of PROSER2 suppressed the invasion and proliferation of cancer cells, while knockdown promoted these processes, suggesting a role in maintaining a less aggressive tumor phenotype. PROSER2 interacts directly with the kinase STK25 and the protein PDCD10, enhancing AMP-activated protein kinase (AMPK) signaling through the STK25-AMPK pathway and contributing to its anti-invasion effects. As such, PROSER2 has been suggested as a potential prognostic marker and possible therapeutic target for certain cancers, although no validated drugs or mechanisms of pharmacological targeting exist to date. Its precise molecular function remains to be fully elucidated, but it is implicated in regulation of cell proliferation, invasion, and signaling pathways associated with tumor progression.
