Overview

Proline and serine-rich protein 3 (PROSER3) is a human protein encoded by the PROSER3 gene (also known as C19orf55), located on chromosome 19q13.12.[2] It is a large protein, notably rich in proline and serine residues, and is predominantly localized to the Golgi apparatus and centrosome, suggesting a role as a centrosome component. PROSER3 is expressed at low levels in various human tissues, with notable variability. Functional studies have linked PROSER3 to centriole biogenesis and microtubule organization. It physically interacts with several centrosomal and cytoskeletal proteins, such as CEP135, NINL, CEP128, CCDC146, and NME7, and is implicated in vesicular trafficking, microtubule dynamics, and cell cycle-related processes. Experimental evidence suggests roles in regulating cell growth and apoptosis, particularly in papillary thyroid cancer cells, where targeting PROSER3 can enhance treatment efficacy and reduce recurrence. Several SNPs in PROSER3 have been described; some overlap with regulatory motifs for RNA-binding proteins and transcription factors, potentially contributing to neurodegenerative and oncogenic processes if mutated[2]. However, there is no evidence that PROSER3 is a direct therapeutic target, nor are there any currently known drugs or biomarker applications associated with it.
