Overview

Proline-containing gluten peptides are short protein fragments derived from the digestion of gluten proteins, especially gliadins, found in wheat and related grains[3][6]. They are notably rich in the amino acid proline, which creates structural kinks making them resistant to breakdown by typical human gastrointestinal proteases[2][3][6]. This resistance allows peptides, such as the highly immunogenic 33-mer from α-gliadin, to persist in the digestive tract and reach the small intestine intact. In genetically susceptible individuals (HLA-DQ2/DQ8 positive), these peptides bind to MHC class II molecules and trigger pathogenic T-cell-mediated immune responses, leading to the characteristic gut inflammation of celiac disease[4][5]. They are not classical therapeutic targets like receptors or enzymes but are the molecular trigger for celiac disease pathogenesis. Enzymatic therapies aimed at degrading these proline-rich peptides are an active area of research for celiac disease management[1][5].
