Overview

PE proteins are a unique class of proteins found primarily in the Mycobacterium tuberculosis complex, characterized by a conserved Proline-Glutamic acid (PE) motif at their N-terminus. They constitute approximately 10% of the M. tuberculosis genome and are typically co-expressed and secreted with PPE (Proline-Proline-Glutamic acid) family proteins via the Type VII (ESX) secretion system. These proteins play a critical role in the pathogenesis of tuberculosis by mediating host-pathogen interactions, facilitating immune evasion through antigenic variation, and modulating host macrophage functions such as phagosome maturation and cytokine signaling. Due to their high immunogenicity and surface localization, PE proteins are major targets for the development of next-generation tuberculosis vaccines and diagnostic tools. Therapeutic strategies also explore inhibiting the secretion machinery (ESX) required for their transport to the bacterial surface, thereby attenuating the pathogen's virulence.
