Overview

Proline rich coiled-coil 2A (PRRC2A) is a large proline-rich protein encoded within the major histocompatibility complex III region and is highly expressed in neural cells, particularly oligodendrocyte precursor cells[1]. It functions as an N6-methyladenosine (m6A) reader protein that binds methylated mRNAs, stabilizing transcripts such as Olig2 to regulate oligodendrocyte specification and myelination by post-transcriptional mechanisms[1][3]. PRRC2A is also involved in translational regulation by promoting leaky scanning and facilitating translation of mRNAs with upstream open reading frames (uORFs)[3]. Loss of PRRC2A impairs nervous system development and is associated with neurodevelopmental defects, while genetic variants and altered expression have been reported in several cancers and autoimmune conditions[3][8]. The protein may also participate in pre-mRNA splicing and is considered essential for optimal cell proliferation[3][4]. No direct evidence supports its use as a therapeutic target, for which reason no specific drugs or mechanisms of action are reported.
